Which constraint can I use if there is a teacher that is sharing a combination of subjects and that must be on the same day.
Example: Ronald teaches Biology, Rodwell teaches IT Practical and Roland teach IT Theory
Biology/IT Practical (3 Periods) is taught at the same time
Biology/IT Theory (2 Periods) is taught at the same time
These students are the same.
Which constraint must l use for these subjects not to be on the same day. Meaning all the 5 are on different days. Monday to Friday.

You are asking same time, but also not same day. I am not sure I understand correctly. For same time, there is a constraint activities same starting time (day+hour), or same starting hour (any days). For not same day, you could use a constraint min 1 days 100% between activities. These are the easiest and most efficient solutions - but other constraints can be used as well, if these cannot be used. Let me know if you need other solutions.
